چکیده

A defined model system which may have relevance to "spontaneous" tumors was described. Cells derived from normal germ-free Wistar rat embryos were propagated in vitro and showed morphologic conversion to epithelium-like cells on the 8th passage. The 19th passage in vitro of the converted cells induced progressively growing local fibrosarcomas in newborn rats; the tumors were not transplantable to additional rats. Sprague-Dawley rats were more receptive to the transformed Wistar cells than Wistar or Fischer rats. The inoculated rats, the normal and transformed cells cultures, and the tumors were free of detectable virus particles and viral antigens. The results indicate that transformation of cells in vitro and their ability to initiate tumors can occur without the involvement of a detectable exotic microbial factor.

Role of Spontaneous Transformation in Carcinogenesis: Development of Preneoplastic Rat TrachéalEpithelial Cells at a Constant Rate

The rate of spontaneous transformation of normal rat trachéal epithe lial (RTE) cells to preneoplastic enhanced growth (EG) variants was estimated in serum-free culture. Spontaneous transformation of RTE cells has previously been observed, but an accurate estimation of the rate of change has not been possible due to the use of serum and feeder cells in the cultures which prevents both unlimite...

Comparison of Germ Cell Gene Expressions in Spontaneous Monolayer versus Embryoid Body Differentiation of Mouse Embryonic Stem Cells toward Germ Cells

Objective Genetic and morphologic similarities between mouse embryonic stem cell (ESCs) and Primordial Germ Cell (PGCs) make it difficult to distinguish the two cell types in in vitro differentiation. Using the expression of specific markers of germ cells that are not expressed or expressed at low levels in ESCs, can help recognizing in vitro differentiated cells MaterialsAndMethods In this stu...

Morphology and serum dependence of cloned cell lines undergoing spontaneous malignant transformation in culture.

A number of morphological changes were found to correlate with the occurrence of spontaneous neoplastic transformation in sublines of five rigidly isolated clones of mouse embryo fibroblasts. These morphological changes included increased cytoplasmic basophilia, reduced spreading of cells on the substrate, increased nuclear:cytoplasmic ratio, greater heterogeneity in the size and shape of cells...

Multistep change in epidermal growth factor receptors during spontaneous neoplastic progression in Chinese hamster embryo fibroblasts.

Whole Chinese hamster embryo lineages have been shown to undergo multistep spontaneous neoplastic progression during serial passage in culture. We have studied the binding, internalization, and degradation of 125I-labeled epidermal growth factor at four different stages of transformation. The whole Chinese hamster embryo cells lost cell surface epidermal growth factor receptors gradually during...
